Not sure at all, if there's a show or movie in particular you want to grab the audio from, try finding it online. If it's from an advertisement, youtube is your friend. As far as grabbing audio from pretty much anything on your computer is concerned, I suggest the program Audio Hijack Pro. It's free and lets you choose which source (program) you want to record from, even add some fx, etc. Great for making samples from movies, etc. to drop into mixes.

You could use a capture card and use software to export the audio out of the video.
Some TV's have an audio output for speakers. Put that output into your soundcard and record with a DAW or sampler. Of course the easy way is to just plug a mic in and record the output of the speakers- and if you have a decent set of speakers and a not-shitty mic, it can sound pretty good (I've done it
