Default Mixing @ my computer desk

If I'm just practicing in my computer room, is it possible to set the main output of the S4 to my desktop computer's speakers? Is there a setting for this anywhere?

You would have to get an adapter to go from your RCA main outs on the S4to the computer speakers. I'm assuming you mean external computer speakers.

RCA out to 1/3 or 1/4 inch TRS

see, that's what I was curious about, and was hoping I could do. I wonder, though, if I'll run into latency issues.

Depends on your pc I suppose.
I've used my Audio2 with the S4 software and it's been fine.

Looks like doing it this way puts your headphone mix through the PC speakers as well...

I've since obtained a couple of tower speakers, a bookshelf speaker and a sub running through a hi fi receiver....works for me!
