I usually drop after I pop it and lock it but thats just me.
on a serious note, you should know when too once you get the feel of it... reading the crowd, energy, etc.

as for the op I and many others will prob say its all about when you feel is right, spur of the moment, I usually have certain songs that need a big drop in so I usually have cue points mapped that 1 will be intro, 2 will be main riff, 3 will be breakdown, 4 will be drop then 5 and 6 will be random different phrases, this way If I know 2 comes in with a pretty funky melody I will do a build up with filter sweep, gater/filter sweep, delay freeze build up, beatmasher/gater, loops etc, then drop when the typical phrase will change...

It depends really, I remember listening to an Eddie Halliwell set, which is also probably my favourite ever set (Godskitchen NYE 2007 if you're interested). The song in question is Bulldozer by Simon PattersonI'm not exactly sure what he does but I presume that he just loops the silence around 5m30s for roughly ten seconds to let the anticipation build up. He then stops the loop and lets it drop, much to the crowds delight.
